چکیده

After the administration of 3Hand 14C-cholesterol to rats 3 days and 4 hr prior to their death respectively, ester and free cholesterol in adrenal glands were successfully labeled separately with different isotopes. Examination of the 3H/14C ratio in free and ester cholesterol of adrenal glands and in plasma corticosterone enabled us to study of the role of ester cholesterol in corticosteroidogenesis in ACTH stimulated adrenal glands. By the ACTH administration, the rate of ester cholesterol hydrolysis was increased, while the rate of esterification of free cholesterol was decreased and the hydrolized cholesterol was incorporated into the free cholesterol pool in mitochondria, and then utilized for corticosterone biosynthesis. The administration of cycloheximide did not interfere with the stimulated rate of ester cholesterol hydrolysis in the ACTH-treated adrenals but it prevented the response of plasma corticosterone to ACTH.

ACTH-induced hydrolysis of cholesteryl esters in rat adrenal cells.

Rat adrenal cortical cells have been prepared by collagenase dissociation of trypsin-treated adrenal tissue. The content and compositions of cholesteryl ester, phospholipid, and triglyceride fatty acids compare favorably with those of undissociated rat adrenal tissue. During 2-hour control incubations of adrenal cortical cells, steroidogenesis was not detected, and the levels of sterol ester, p...

Failure of adrenal corticosterone production in POMC-deficient mice results from lack of integrated effects of POMC peptides on multiple factors.

Production of corticosteroids from the adrenal gland is a multistep process in which corticosterone is enzymatically processed from its precursor cholesterol. The main hormone regulating the production of corticosterone is the proopiomelanocortin (POMC)-derived adrenocorticotropic hormone (ACTH). Adrenals of POMC-deficient (POMC(-/-)) mice do not produce corticosterone either at basal levels or...

Investigation of the relation between the hypothalamus-pituitary-adrenal (HPA) axis activity, IL-6 and hyperalgesia during chronic inflammation due to rheumatoid arthritis (RA) in male rats.

Introduction: The activity of Hypothalamus-Pituitary-Adrenal (HPA) axis is increased following inflammation due to its closed relation with immune system. This axis indicates an increased secretion of ACTH, and corticosterone during acute inflammation while little is known about its activity during chronic inflammation such as rheumatoid arthritis (RA). In this study we measured the products...

Adrenocortical response to corticotropin is potentiated by part of the amino-terminal region of pro-corticotropin/endorphin.

Five peptides derived from pro-corticotropin/endorphin (pro-ACTH/endorphin), the pituitary corticotroph cell prohormone, were bioassayed with isolated rat adrenocortical cells: alpha- and beta-melanotropin, beta-lipotropin, beta-endorphin, and the amino-terminal region of pro-ACTH/endorphin known as "16k fragment." The effect of each on steroidogenesis was measured at potentially physiological ...

Relative importance of high and low density lipoproteins in the regulation of cholesterol synthesis in the adrenal gland, ovary, and testis of the rat.

These studies were undertaken to determine the relative importance of plasma lipoproteins and trophic hormones as regulators of cholesterol synthesis in three endocrine glands of the rat. In rats treated with 4-aminopyrazolo[3,4-dlpyrimidine, the plasma cholesterol level was reduced from 56 to 5 mg/dl and this resulted in a decrease in cholesterol ester content in the adrenal gland from 26.26 t...
